當脂酶的活性被抑制後，食物中的三酸甘油脂就不會被分解成可吸收的脂肪酸形式，反而會以脂肪形式被排出去。因此減肥藥奧利司他的主要作用是抑制脂酶，並使脂肪無法被吸收而最後隨著糞便排出。
